I don’t get it

i think the joke is actually that they’re all realizing they’re trans at the same time since they’re all “peter” parkers.
I prefer your interpretation and rescind my own.
Nah, it’s about them being trans in one universe so possibly more of them might also be. “You girls can figure this out on your own” is referring to all of them as girls.
The non-Peter spiders are amused by this.

Maybe you were thinking of Doc Ock…? He becomes Spider-Man in an alternate universe.
